geldanamycin(English)

  • a member of the benzoquinone ansamycin family of antibiotics, with isolation first reported in 1970. Has the ability to cause transformed cells to revert to normal phenotypes, but its molecular target, heat shock protein 90 (Hsp90), was identified only in the mid-1990s. Geldanamycin binds to Hsp90, preventing its ATPase activity and ultimately its ability to stabilize key proteins involved in cell cycle regulation, apoptosis, oncogenesis, and cell growth, thereby resulting in a multipronged attack on cancer-causing processes
